Isotype controls are negative controls that are designed to measure the amount of non-specific background signal caused by the primary antibody. Isotype controls have no specificity for the target protein, yet retain all of the non-specific characteristics of an antibody.
Flow cytometric analysis of anti-CD19 CAR expression on HEK293T cells. HEK293T cells were lentivirally transduced with anti-CD19 CAR and cultured for 3 days. 1×106 cells were stained for the expression of anti-CD19 CAR with Biotin Rabbit IgG Isotype Control (Cat. No.700317, left panel) and Rabbit Anti-Mouse FMC63 scFv Polyclonal Antibody, Biotin (Cat. No. 500014, right panel). Secondary staining was carried out with Streptavidin PE (Cat. No. 700032). Acquisition of >10, 000 events was performed.
Preparation and Storage
Shipped at 2-8°C. Store at 2-8°C short term (1-2 weeks). Store at -20°C in small aliquots for long term storage. Avoid freeze/thaw cycle.
The polyclonal antibody was purified by Protein A.
The antibody was conjugated with biotin under optimum conditions, and unreacted biotin was removed.
1. Dilute this control antibody to the same concentration (not dilution) as the specific antibody used for analysis.
2. Biotinylated antibodies are designed to be detected using streptavidin conjugates.
3. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
